Description | Interferon gamma (IFN-gamma) is produced by cells that are involved in inflammatory immune responses. IFN-gamma is predominantly released by memory and effector CD4+ and CD8+ T cells as well as by NK cells upon activation. Quantitative analysis of IFN-gamma–producing cells can provide important information on the course of immune responses. Anti-IFN‑gamma antibodies are designed for intracellular staining of IFN‑gamma–producing cells. Cells can be stimulated for IFN-gamma–production, e.g. by polyclonal stimulation with mitogens. For induction of IFN‑gamma–production by antigen-specific T cells, cells are restimulated with respective antigen. IFN‑gamma can be accumulated in the cells by addition of secretion inhibitors like brefeldin A. After fixation and permeabilization of the cell sample, IFN‑gamma–producing cells can be stained intracellularly with Anti-IFN‑gamma antibodies. Staining of surface markers allows simultaneous flow cytometric analysis of subsets and activation status of the IFN-gamma–producing cells. | |

About IFN-gamma
This gene encodes a soluble cytokine that is a member of the type II interferon class. The encoded protein is secreted by cells of both the innate and adaptive immune systems. The active protein is a homodimer that binds to the interferon gamma receptor which triggers a cellular response to viral and microbial infections. APC-Vio®770 from Miltenyi is a tandem fluorophore with APC as the donor and has an excitation peak at 652 nm and an emission peak at 775 nm, and is spectrally similar to APC-Cy® 7 (GE Healthcare) APC-H7 (BD Biosciences), APC-eFluor™ 780 (ThermoFisher Scientific), APC-Vio®770 (Miltenyi Biotec), and APC-Fire™ 750 (BioLegend). The Vio® dye family are products of Miltenyi Biotec, with many antibody conjugates designed and validated for flow cytometry.
