Description CIV1.1 may be used for the affinity purification of the Anaphase Promoting Complex (APC/C) by peptide elution. Suggested Dilution for western blot: 1:500. It is suggested to blot for at least 2 hours (wet transfer) to ensure that APC4 is transferred efficiently.Mass spectrometry was used to confirm that the whole APC/C is co-precipitated. To establish the APC4 specific bands, it is suggested to do an APC4 immunoprecipitation assay showing input, IP and FT and blot for at least 2 hours.

About Apc4
A large protein complex, termed the anaphase-promoting complex (APC), or the cyclosome, promotes metaphase-anaphase transition by ubiquitinating its specific substrates such as mitotic cyclins and anaphase inhibitor, which are subsequently degraded by the 26S proteasome. Biochemical studies have shown that the vertebrate APC contains eight subunits. The composition of the APC is highly conserved in organisms from yeast to humans. The exact function of this gene product is not known. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Nov 2013]
About DyLight 650
DyLight™ 650 is a red-emitting fluorophore that excited by the 640 nm laser and collected using a 670/30 nm bandpass filter. DyLight™ 650 has an excitation peak at 652 nm and an emission peak at 672 nm, and is spectrally similar to Alexa Fluor™ 647 and Cy5. DyLight™ 650 is most commonly used in flow cytometery and fluorescence microscopy applications.
