Description Clone REA160 recognizes the human linker for activation of T cell family member 1 (LAT) antigen phosphorylated at tyrosine 171 (pY171). LAT is an integral membrane protein that was initially detected as a major PTK substrate during T cell–receptor (TCR) activation. It is also involved in low affinity immunoglobulin gamma Fc region receptor III mediated signaling in natural killer cells and high affinity immunoglobulin epsilon receptor mediated signaling in mast cells. TCR-mediated LAT phosphorylation is critical for the membrane recruitment of signaling complexes required for T cell–activation. The LAT protein contains a short extracellular domain, a transmembrane sequence, and an extended cytoplasmic domain with nine tyrosine residues conserved in the mouse and human sequences. Some of these tyrosine residues have been shown to be docking sites for mediators of TCR signaling. Tyrosine 171 is suggested to be involved in more than merely creating SH2-binding sites and may affect the native conformation, localization, stability, and/or sequential phosphorylation of LAT during TCR stimulation. | Additional information: Clone REA160 displays negligible binding to Fc receptors.
