Description | Indo-1 acetomethoxy (AM) is a popular UV-excitable calcium indicator, similar to Fura-2. In contrast to Fura-2, Indo-1 has a dual emission peak. The main emission peak in calcium-free solution is 475nm while in the presence of calcium the emission is shifted to 400nm. Indo-1 AM is a membrane permeable probe and can be easily loaded into cells by passive incubation. The probe is widely used in flow cytometry. About Indo-Lo (Ca2+free)
Indo-1, AM is a ratiometric calcium indicator optimally excited by a 355nm UV laser or excitation source. After Indo-1, AM is loaded into live cells, it is hydrolyzed and available to bind cytoplasmic Ca2+. Probe bound to Ca2+ emit at ~400nm, while probe unbound to Ca2+ emits at ~475nm. Because of these unique spectral characteristics, it's the preferred calcium indicator for flow cytometry and is often frequently used in fluorescence live-cell microscopy.
