Description Clone REA185 recognizes CD42b, a 145 kDa single-pass type I membrane protein. Expression of CD42b is found on platelets and megakaryocytes. CD42b, the alpha chain, together with the beta chain (CD42c), forms the heterodimer, glycoprotein Ib (GP Ib). The GP Ib receptor complex further includes association of the alpha and beta subunits with platelet glycoprotein IX and glycoprotein V and this complex serves as the receptor for von Willebrand factor (VWF). The interaction of the GP Ib-IX-V complex to VWF initiates initial platelet adhesion to vascular subendothelium after vascular injury, platelet activation, thrombosis, and hemostasis. Other known ligands for GPIb–IX–V include alpha-thrombin, clotting factors XI/XIIa, and high-molecularweight kininogen. | Additional information: Clone REA185 displays negligible binding to Fc receptors. |

About CD42b
Glycoprotein Ib (GP Ib) is a platelet surface membrane glycoprotein composed of a heterodimer, an alpha chain and a beta chain, that is linked by disulfide bonds. The Gp Ib functions as a receptor for von Willebrand factor (VWF). The complete receptor complex includes noncovalent association of the alpha and beta subunits with platelet glycoprotein IX and platelet glycoprotein V. The binding of the GP Ib-IX-V complex to VWF facilitates initial platelet adhesion to vascular subendothelium after vascular injury, and also initiates signaling events within the platelet that lead to enhanced platelet activation, thrombosis, and hemostasis. This gene encodes the alpha subunit. Mutations in this gene result in Bernard-Soulier syndromes and platelet-type von Willebrand disease. The coding region of this gene is known to contain a polymophic variable number tandem repeat (VNTR) domain that is associated with susceptibility to nonarteritic anterior ischemic optic neuropathy. About PE-Vio770
PE-Vio® 770 from Miltenyi Biotec is a red-emitting tandem fluorophore that combines pycoerythrin (PE) and Vio®770. The donor molecule, PE can be excited by the 488-nm blue, 532-nm green, or 561-nm yellow-green laser and and transfers energy to the acceptor molecule, Vio®770, which emitts light that can be captured with a 780/60 nm bandpass filter. PE-Vio®770 has an excitation peak at 565 nm and an emission peak at 775 nm and is a common alternative to PE-Cy7 and PE-H7.
