Description Clone REA319 recognizes the human CD162 antigen, a single-pass type I membrane protein which is also known as P-selectin glycoprotein ligand 1 (PSGL-1). CD162 is expressed on neutrophils, monocytes, and most lymphocytes. It is the high affinity receptor for CD62P (P-selectin) on myeloid cells and stimulated T lymphocytes and mediates rapid rolling of leukocytes over vascular surfaces during the initial steps in inflammation. CD162 is also involved in homeostasis including stem cell homing to the thymus and mature T cell homing to secondary lymphoid organs. It has been found to bind the homeostatic chemokines CCL19 and CCL21 and to support the chemotactic response to these chemokines. | Additional information: Clone REA319 displays negligible binding to Fc receptors. | |

About CD162
This gene encodes a glycoprotein that functions as a high affinity counter-receptor for the cell adhesion molecules P-, E- and L- selectin expressed on myeloid cells and stimulated T lymphocytes. As such, this protein plays a critical role in leukocyte trafficking during inflammation by tethering of leukocytes to activated platelets or endothelia expressing selectins. This protein requires two post-translational modifications, tyrosine sulfation and the addition of the sialyl Lewis x tetrasaccharide (sLex) to its O-linked glycans, for its high-affinity binding activity. Aberrant expression of this gene and polymorphisms in this gene are associated with defects in the innate and adaptive immune response. Alternate splicing results in multiple transcript variants.[provided by RefSeq, Apr 2011]
About FITC
Fluorescein isothiocyanate (FITC) has an excitation peak at 495 nm and an emission peak at 519 nm. The name FITC is a misnomer in that the isothiocyanate is a reactive form of this dye. Once FITC is conjugated to an antibody, it is simply Fluorescein conjugated. FITC is one of the most widely used dyes for fluorescent applications, therefore most instruments come standard with a 488 nm laser and FITC filter set up. FITC is commonly conjugated to secondary antibodies and used in applications such as flow cytometry, immunocytochemistry, and immunohistochemistry. FITC is relatively dim, sensitive to photobleaching and it is susceptible to changes is pH. There are better performing alternatives to FITC, like Vio®Bright 515, Alexa Fluor™ 488, iFluor® 488, CF®488A and DY-488. FITC is a long-time generic dye with no sole manufacturer or trademark.
