Description Clone REA441 recognizes the human and mouse integrin beta7 antigen, a single-pass type I membrane protein which is also known as gut homing receptor beta subunit. Integrins are heterodimeric glycoprotein receptors and exist as non-covalently bound alpha and beta subunits. The integrin beta7 subfamily has two known members (alpha4beta7 and alphaEbeta7) that are expressed primarily by leukocytes and have been implicated in secondary lymphoid structure formation, plasma cells homing to gastrointestinal mucosa, and inflammatory responses. Their ligands: the mucosal vascular addressin cell adhesion molecule 1 (MAdCAM-1) and E-cadherin are present on the surface of endothelial cells, bone marrow stromal cells, and epithelial cells. Interaction of alpha4beta7 with MAdCAM-1 allows for tissue-specific migration of circulating lymphocytes into the lamina propria and Peyer patches of the gut, whereas alphaEbeta7 retains intraepithelial lymphocytes within the gut epithelium through E-cadherin binding. Integrin beta7 is also reported to be involved in the pathogenesis of several diseases such as colitis, diabetic insulitis, and lymphoid malignancies including lymphomatous polyposis in mantle cell lymphoma, thymic lymphoma, and mucosa-associated T cell and B cell non-Hodgkin lymphomas. | Additional information: Clone REA441 displays negligible binding to Fc receptors.

About Integrin beta 7
This gene encodes a protein that is a member of the integrin superfamily. Members of this family are adhesion receptors that function in signaling from the extracellular matrix to the cell. Integrins are heterodimeric integral membrane proteins composed of an alpha chain and a beta chain. The encoded protein forms dimers with an alpha4 chain or an alphaE chain and plays a role in leukocyte adhesion. Dimerization with alpha4 forms a homing receptor for migration of lymphocytes to the intestinal mucosa and Peyer's patches. Dimerization with alphaE permits binding to the ligand epithelial cadherin, a calcium-dependent adhesion molecule. Alternate splicing results in multiple transcript variants. Additional alternatively spliced transcript variants of this gene have been described, but their full-length nature is not known. [provided by RefSeq, Sep 2013]
About PE-Vio770
PE-Vio® 770 from Miltenyi Biotec is a red-emitting tandem fluorophore that combines pycoerythrin (PE) and Vio®770. The donor molecule, PE can be excited by the 488-nm blue, 532-nm green, or 561-nm yellow-green laser and and transfers energy to the acceptor molecule, Vio®770, which emitts light that can be captured with a 780/60 nm bandpass filter. PE-Vio®770 has an excitation peak at 565 nm and an emission peak at 775 nm and is a common alternative to PE-Cy7 and PE-H7.
