HLA B12 Monoclonal / PE-Vio770 / REA138

Product Details
Description Clone REA138 recognizes HLA B12, a class I human leukocyte antigen (HLA). Expressed on the surface of most nucleated cells, class I molecules are heterodimeric molecules and consist of a type I integral membrane alpha heavy chain and soluble beta2 microglobulin protein. The extracellular region of the heavy chain further consists of three domains, of which one comprises the peptide binding groove. Antigens binding to class I molecules are 8–10 amino acids long and play an important role in recognition of the virus infected and malignant cells by cytotoxic T lymphocytes (CTLs). In addition, class I molecules interact with NK cell receptors to modulate the activity of NK cells. | Additional information: Clone REA138 displays negligible binding to Fc receptors.

About HLA B12
HLA-B belongs to the HLA class I heavy chain paralogues. This class I molecule is a heterodimer consisting of a heavy chain and a light chain (beta-2 microglobulin). The heavy chain is anchored in the membrane. Class I molecules play a central role in the immune system by presenting peptides derived from the endoplasmic reticulum lumen. They are expressed in nearly all cells. The heavy chain is approximately 45 kDa and its gene contains 8 exons. Exon 1 encodes the leader peptide, exon 2 and 3 encode the alpha1 and alpha2 domains, which both bind the peptide, exon 4 encodes the alpha3 domain, exon 5 encodes the transmembrane region and exons 6 and 7 encode the cytoplasmic tail. Polymorphisms within exon 2 and exon 3 are responsible for the peptide binding specificity of each class one molecule. Typing for these polymorphisms is routinely done for bone marrow and kidney transplantation. Hundreds of HLA-B alleles have been described. [provided by RefSeq, Jul 2008]
About PE-Vio770
PE-Vio® 770 from Miltenyi Biotec is a red-emitting tandem fluorophore that combines pycoerythrin (PE) and Vio®770. The donor molecule, PE can be excited by the 488-nm blue, 532-nm green, or 561-nm yellow-green laser and and transfers energy to the acceptor molecule, Vio®770, which emitts light that can be captured with a 780/60 nm bandpass filter. PE-Vio®770 has an excitation peak at 565 nm and an emission peak at 775 nm and is a common alternative to PE-Cy7 and PE-H7.
