Description The monoclonal antibody GHI/61.1 reacts with human CD163, a single-chain transmembrane protein also known as hemoglobin scavenger receptor or M130. It is expressed by mature tissue macrophages and peripheral blood monocytes. The expression of CD163 is up-regulated in vitro and in vivo by anti-inflammatory mediators such as IL-10 and (gluco)corticosteroid and is shed to blood upon inflammatory activation of macrophages. CD163 functions as a high affinity scavenger receptor for the complex of haemoglobin and haptoglobin. Depending on the ligand, crosslinking of M130 initiates signal transduction leading to the production of proinflammatory cytokines Il-1ß, IL-6, and GM-CSF or the antiinflammatory cytokine IL-10.

About CD163
The protein encoded by this gene is a member of the scavenger receptor cysteine-rich (SRCR) superfamily, and is exclusively expressed in monocytes and macrophages. It functions as an acute phase-regulated receptor involved in the clearance and endocytosis of hemoglobin/haptoglobin complexes by macrophages, and may thereby protect tissues from free hemoglobin-mediated oxidative damage. This protein may also function as an innate immune sensor for bacteria and inducer of local inflammation. Alternatively spliced transcript variants encoding different isoforms have been described for this gene. [provided by RefSeq, Aug 2011]
About PE-Vio770
PE-Vio® 770 from Miltenyi Biotec is a red-emitting tandem fluorophore that combines pycoerythrin (PE) and Vio®770. The donor molecule, PE can be excited by the 488-nm blue, 532-nm green, or 561-nm yellow-green laser and and transfers energy to the acceptor molecule, Vio®770, which emitts light that can be captured with a 780/60 nm bandpass filter. PE-Vio®770 has an excitation peak at 565 nm and an emission peak at 775 nm and is a common alternative to PE-Cy7 and PE-H7.
