Description | Tumor necrosis factor alpha (TNF-alpha) is produced by cells that are involved in inflammatory immune responses. TNF-alpha is secreted by activated CD4+ T cells, monocytes, macrophages, NK cells, and neutrophils. | Anti-TNF-alpha antibodies have been designed for intracellular staining of TNF-alpha–producing cells. Cells can be stimulated for TNF-alpha production, for example, by polyclonal stimulation with mitogens. For induction of TNF-alpha production by antigen-specific T cells, cells are restimulated with the respective antigen. TNF-alpha can be accumulated in the cells by addition of secretion inhibitors like brefeldin A. After fixation and permeabilization of the cell sample, TNF-alpha–producing cells can be stained intracellularly with Anti-TNF-alpha antibodies. Staining for surface markers allows simultaneous flow cytometric analysis of subsets and activation status of the TNF-alpha–producing cells. | |

About PE-Vio770
PE-Vio® 770 from Miltenyi Biotec is a red-emitting tandem fluorophore that combines pycoerythrin (PE) and Vio®770. The donor molecule, PE can be excited by the 488-nm blue, 532-nm green, or 561-nm yellow-green laser and and transfers energy to the acceptor molecule, Vio®770, which emitts light that can be captured with a 780/60 nm bandpass filter. PE-Vio®770 has an excitation peak at 565 nm and an emission peak at 775 nm and is a common alternative to PE-Cy7 and PE-H7.
